Biotechnological and Biological Product Analysis
Intertek ASG has experience of providing full analytical packages or individual analyses. Analyses are applicable from early through to late stage product development programmes, and typical examples have included the following analytical technology areas:
- Determination of protein intact molecular weight by mass spectrometry
- Peptide Mapping (fingerprinting) including N- and C-terminal sequencing by mass spectrometry
- Amino acid composition including determination of the extinction coefficient
- Electrophoretic and isoform patterns determined by isoelectric focusing and SDS-PAGE
- Liquid chromatographic analysis using suitable chromatographic procedures
- N-terminal Sequencing (using Edman Chemistry)
- Sufhydryl group and disulfide bridge assignment by MS and MS/MS analysis and sites of mismatched disulfide (-S-S-) forms
- Determination of carbohydrate content (neutral sugars, amino sugars, sialic acids)
- Analysis of the structure of carbohydrate chains, oligosaccharide patterns and the glycosylation sites on the polypeptide chain
- Identification of other post-translational modifications (PTMs, e.g. acetylation, phosphorylation, addition of lipids)
- Product-related impurities including degradation products (e.g. de-amidation, acetylation, oxidation, or carbamylation)
